PI3K (p110alpha (E542K)/p85alpha)
The PI3K (p110alpha (E542K)/p85alpha) Kinase Enzyme Activity Assay Kit offers a validated, ready-to-use biochemical platform for accurately measuring kinase inhibitor efficacy targeting the PI3K complex. The kit includes recombinant PI3K (p110α (E542K)//p85α), substrate, ATP, detection reagents, and assay plates—enabling precise quantification of ADP accumulation across two, six or twenty five 384-well plate formats. Designed to handle elevated ATP concentrations, this assay delivers robust, reliable data ideal for high-throughput screening (HTS) and other drug discovery applications.
Biological information
The PI3K (p110α (E542K)/p85α) Kinase
Activity Assay Kit is a robust biochemical tool
designed to evaluate the efficacy of kinase
inhibitors. Utilizing advanced assay
technology, it has been specifically
optimized for the PI3K (p110α
(E542K)/p85α) complex. All necessary
reagents are included—such as the recombinant PI3K (p110α (E542K)/p85α)
complex (Eurofins #14-782), substrate, ATP,
detection reagents, and assay plates—to
ensure a complete, ready-to-use solution. A
Certificate of Analysis (COA) accompanies
the kit, detailing the optimal reaction
parameters for detecting kinase inhibitors.
The kit provides sufficient material for
running assays on two, six or twenty five
384-well plate formats.
The E542K substitution is a somatic
mutation in p110α linked to tumors of the
colon and brain. Combined in vitro and in
vivo studies have demonstrated that this
mutation leads to enhanced lipid kinase
activity compared to the wild type and is
capable of inducing oncogenic
transformation (Kang S. et al., PNAS, 2005;
102:802-807 and Zhao J.J. et al., PNAS, 2005;
102:18443-18448).
