Background
Phosphoinositide-Dependent Kinase-1 (PDK1) plays a central role in mediating cell signaling between PI3K and various intracellular serine/threonine kinases by phosphorylating many other kinases including PKC, p70S6 kinase, ribosomal S6 kinase, p21-activated kinase, PKC-related kinase, PKB and serum glucocorticoid kinase. PDK1 interacts with a wide range of substrates, and has been found to regulate apoptosis, cell proliferation and survival through the PI3K signaling pathway. A PDK1 inhibitor may be of therapeutic use in the treatment of human cancers and diabetes.
