Background
Checkpoint kinase (CHK) is activated by a broad spectrum of DNA-damaging agents, including DNA-strand-breaking agents such as ionizing radiation and topoisomerase inhibitors, and those agents that cause replication stress such as ultraviolet light, hydroxyurea and 5-fluorouracil. CHK1 regulates numerous checkpoint pathways, including S-phase and G2-M checkpoints. Other potential CHK1 functions include replication, chromatin remodelling and DNA repair. CHK1 inhibitors may sensitize tumors to chemotherapy and radiotherapy.
