It comes with all essential components, including recombinant MSK2 (Eurofins #14-616), a substrate, ATP, detection reagents, and assay plates. Each kit is accompanied by a Certificate of Analysis (COA), which outlines the recommended reaction conditions for accurate inhibitor detection. The supplied materials are sufficient to perform experiments covering two, six or twenty five 384-well assay plates.

MSK2 is a mitogen- and stress-activated serine/threonine kinase that works together with MSK1 to couple MAPK signalling to transcriptional and chromatin responses. Mechanistically, MSK2 is activated downstream of ERK1/2 and p38 MAPKs and mediates phosphorylation of transcription factors (e.g. CREB1, ATF1) and chromatin substrates (notably histone H3), thereby regulating stimulus-dependent gene expression. In lipopolysaccharide-stimulated primary macrophages, MSK2 acts downstream of the Toll-like receptor TLR4 to limit the production of pro-inflammatory cytokines.

MSK2 is upregulated in certain cancers, including cervical cancer, and has been shown to promote proliferation and tumour formation.
