Background
NSD1 (Nuclear receptor-binding SET domain protein 1) is a histone methyltransferase that belongs to the SET domain-containing protein family. NSD1 specifically targets histone H3 at lysine 36 (H3K36), and catalyzes its mono-, di-, and tri-methylation, leading to changes in chromatin structure and gene expression. NSD1 has been implicated in the regulation of development and growth, including the regulation of body size and brain development. It has also been shown to play a role in the development of several types of cancer, including leukemia and solid tumors. In addition, mutations in the NSD1 gene have been associated with Sotos syndrome, a rare genetic disorder characterized by overgrowth, developmental delay, and intellectual disability. NSD1 is an important epigenetic protein with potential therapeutic applications. Its inhibition has been shown to suppress tumor growth and improve response to chemotherapy in various types of cancer.
