Background
Purified using glutathione sepharose followed by gel filtration. Combined in vitro with CK1δ, active (Item # 14-520). MDM2 (Mouse Double Minute 2) is an E3 ubiquitin ligase that targets the tumor suppressor protein p53 for degradation by the proteasome. CK1δ (Casein Kinase 1 Delta) is a protein kinase that can phosphorylate MDM2. MDM2 can also be ubiquitinated by other E3 ubiquitin ligases, such as CHIP (C-terminus of Hsc70-Interacting Protein) and RNF5 (Ring Finger Protein 5), which can lead to MDM2 degradation. However, CK1δ-mediated phosphorylation of MDM2 has been shown to prevent its degradation by CHIP and RNF5, leading to increased stability of MDM2. Overall, the ubiquitination of MDM2 by CK1δ and other E3 ubiquitin ligases plays a crucial role in regulating the stability and activity of MDM2 and p53, which are important players in tumor suppression and cell survival.
