A collaborative gene-to-structure workflow using cryoEM for membrane protein targets

CALIXAR is proud to present its collaborative project with NanoImaging Services, focused on fully native membrane protein structure determination by cryoEM.

Cryogenic electron microscopy (cryoEM) is a powerful method for high-resolution three-dimensional (3D) protein reconstruction. To perform such a characterization the high-quality and stable samples for high-resolution protein reconstruction, is essential to reduce project running times.

In this project, CALIXAR brought its expertise in fully native membrane protein preparation for more than 10 years. The combination of cryoEM technologies and expertise in protein sample preparation is returning protein structures at higher resolutions in shorter timescales.

This project combined CALIXAR’s production of the fully native AcrB target from gene to highly pure protein, with up to 100% solubilization and purification using CALIXAR’s proprietary detergents increasing by 16% the thermostability of the protein. Thanks to its experience in cryoEM, NanoImaging Services performed 3D reconstruction of AcrB in 10 days.

The association of NanoImaging Services & CALIXAR expertises is enabling Pharmas, Biotechs, and CROs to perform structural analysis of high-value drug targets in short amounts of time without having to purchase cryoEM equipment or recruiting experts in protein sample optimization, while accessing to cutting-edge technologies.
